INgezim PPV está basado en la técnica de ELISA indirecto que utiliza un anticuerpo monoclonal (AcM) específico de
inmunoglobulinas porcinas (Igs), y antígeno recombinante (proteína VP2 de PPV).
BASE TÉCNICA DEL KIT
1. Las placas se suministran tapizadas con antígeno de PPV (cápsidas
recombinantes de proteína VP2). Las muestras de suero se añaden en los
pocillos y se incuban.
2. Si las muestras contienen anticuerpos específicos de PPV, éstos se unirán al
antígeno.
3. Cuando se añade un AcM-PO específico de inmunoglobulinas porcinas, éste
se unirá a las Igs unidas al antígeno. Esta unión se revela mediante reacción
colorimétrica tras adición de substrato.

El ensayo establece un Cut Off. Las muestras con un valor de DO superior al
Cut Off se consideran Positivas, y las muestras con un valor de DO inferior al
Cut Off se consideran Negativas.
El título de la muestra será la última dilución de la misma que presente un valor
de DO mayor que el cut off.

SENSIBILIDAD Y ESPECIFICIDAD
1. Correspondencia con la técnica de referencia IHA
(Inhibición de la Hemaglutinación)
Con objeto de determinar la sensibilidad y especificidad de
INgezim respecto a la técnica IHA, se analizaron 238 sueros
de animales de campo.
3. Sensibilidad utilizando sueros de animales vacunados
experimentalmente
A partir de animales vacunados experimentalmente, se
obtuvo el perfil de detección de anticuerpos de este ensayo.
En el mismo estudio se llevó a cabo una comparación con la
técnica IHA. Los resultados obtenidos se muestran en la
siguiente figura.

2. Sensibilidad analítica
En la segunda semana post-infección, el ensayo puede
detectar títulos de anticuerpos en torno a 1/200.

4. Especificidad utilizando animales SPF
Con objeto de determinar la especificidad del ensayo, se
utilizó un panel de 188 animales negativos SPF. Los
resultados obtenidos indicaron que la especificidad del
ensayo era del 100%.

5. Titulación a pocillo único
Es posible obtener un título aproximado del suero basándose
en el valor de su DO, mediante el uso de una fórmula que
corresponde a una curva exponencial.

PPV is based on an indirect ELISA technique, which uses a monoclonal antibody (MAb) specific to porcine
immunoglobulines (Igs), and recombinant antigen (VP2 protein of PPV).

CONJUGATE ADDITION
Plates are coated with PPV antigen (recombinant capsides of VP2). Sera
samples are added and incubated.
If the samples contain antibodies specific to PPV, they will bind to the
antigen.
When a MAb-PO specific of porcine immunoglobulines is added, it will
bind to the Igs bound to the antigen. The binding is detected by the
development of a colorimetric reaction after the addition of the
substrate.

Detection and /or titration of specific antibodies to PPV in porcine sera
samples.
WASHING
INTERPRETATION OF THE RESULTS
There is one Cut Off for the results interpretation. Samples with an OD higher
than the Cut Off value are positive and samples with an OD lower than
theCut Off value are negative. The titter of the sample will be the last dilution
of the sample showing an OD higher than the Cut Off value.

238 field sera were analysed, to determine the sensitivity
and the specificity of INGEZIM PPV ELISA technique in
relation to the IHA technique
3. Sensitivity using experimentally vaccinated sera.
Experimentally vaccinated animals were used and the
antibodies detection profile was obtained. In the same
study, a comparison with the IHA results was made. The
results are represented in the following figure.

In order to determine the specificity of the assay, a set of

5. One well titration of sera
It is possible to obtain an approximated titre of the sera
based on its OD value, by using a formula which
corresponds with an exponential curve.
